The driver of the car, a 25-year-old woman, and the six-year-old boy, were airlifted from the scene after being freed from the wreckage.
Emergency services were called to the scene just before 10am.
Duty officer with Chifley Local Area Command, Inspector David Abercrombie, said initial investigations were still underway and it was too early to determine what caused the accident.
Inspector Abercrombie said the silver Ford Fiesta was travelling towards Bathurst when it appears the driver lost control of the car which collided heavily with the front of the prime mover, which was driving towards Mudgee.
As a result the driver and the passenger were trapped in the car for a short time, before being airlifted from the scene.
The driver, a 25-year-old woman suffered multiple injuries, including fractures, particularly to her legs.
Crash investigators remain at the scene, and traffic diversions are in place.
