Held in conjunction with the Perthville Fair, which will be held nearby on the same day, the tours will explore the rooms of the original convent built in November, 1872.
The tour is set to begin in the Founders Room, where a painting of Julian Tenison Woods and Mary MacKillop by Reg Campbell hangs.
Sister Mary Murphy said the Founders Room has served difference purposes over the years.
"This used to be the sisters sitting room and an office as well over the years," she said.
Visitors will also see a detailed wall panel that features of the names of the sisters who have been laid to rest at the Perthville convent, with the year of the death and age included.
The history room will also be opened, which has detailed timelines of Julian Tenison Woods and Mary MacKillop's lives.
Old photos also show how the convent has grown in the past 147 years, as well as the values of Fr Woods and Sr MacKillop displayed on a plaque.
People will also be able to spend quiet time in the historic St Joseph Chapel, which was opened in 1960.
The tour cost is $5 per person, starting at 11am and 1pm sharp, lasting approximately one hour in duration.
No booking is required but people are asked to meet the tour guide at the front of the St Joseph Administration Centre with 10 minutes before the tour start time.
