The five-day event, starting in Tamworth on Wednesday, will see the C3 Cyclists attempt to raise $75,000 for the Humpty Dumpty Foundation.
C3 Cyclists president Steve Dunstall said the funds will go towards lifesaving medical equipment for children in various NSW hospitals, including Bathurst Base.
"Certain children and their families have to travel great distances to receive lifesaving treatment, so we're aiming to fund the equipment necessary to ensure they can be treated locally," Mr Dunstall said.
"We raised around $50,000 each from the previous two events, and we're hoping to raise the bar this year."

The fundraising will aim to secure four Airvo 2 humidifiers [valued at $2950 each] for Bathurst Base Hospital's emergency department and paedriatric unit.
C3 Cyclists are also intending to fund medical equipment for hospitals in Tamworth, Maitland and Newcastle [John Hunter Children's Hospital].
"The Humpty Dumpty Foundation purchases the equipment for the hospital, and they do an incredible job in saving children's lives," Mr Dunstall said.
"Some of our riders have been personally affected with their own kids needing specialist medical care, and they only know too well the importance of having the right equipment where it is needed."
VERTO chief executive officer Ron Maxwell said it was critical for the organisation to assist the C3 team in meeting their goals.
"We've been involved for three years now, and it's fantastic to see this fundraising initiative benefit sick children," Mr Maxwell said.
"Part of VERTO's charter is to give back to the community we operate in, and this is helping change people's lives."
VERTO's president of the board Andrew Abel has been involved in the event for four years, and said it's a practical opportunity to visit the communities who will benefit from the initiative.
"It's a powerful thing when you can get out to the hospitals, speak to the medical personnel and hand over pieces of equipment that will make a difference to the lives of children," Mr Abel said.
"We've already raised $20,000, and we expect to send that total north of $50,000."
Since 2010, C3 Cyclists has raised over $275,000 for the Humpty Dumpty Foundation.
