The bookstore was informed of its win on Tuesday, after last week's awards ceremony in Queensland was cancelled due to the coronavirus.
This year's award goes alongside the other bookstore of the year awards won by the store in 2010, 2013 and 2016.
BooksPlus owner Kathryn Bancroft said it was fantastic to be acknowledged.
"We couldn't do what we do without the community's support," she said.
BooksPlus was judged on their success and commitment to excellence in book retailing, community engagement and creative marketing.
BooksPlus was a finalist in the regional category, alongside another two bookstores - Megalong Books in the Blue Mountains and Paperbark Merchants in Albany, Western Australia.
The regional category is for bookstores that are not in the metro area, which has its own award.
"To be acknowledged by your industry is great. It's like winning a Walkey in journalism," she said.
While the Sydney events have been cancelled, Ms Bancroft said the Bathurst Readers' and Writers' Festival is hopeful of continuing with events online.
