Premier Gladys Berejiklian announced the extension in the 11am press conference, saying the situation in western NSW, particularly in Dubbo, was of great concern.
"We do appreciate there are many communities in the regions that don't have any cases, that have never had any cases and they're really wanting to come out of lockdown, but as a precaution the health advice says that we should extend the lockdown to all of regional NSW," she said.
Deputy premier John Barilaro added that the regions were "sitting on a knife edge", and the extension was essential to protect communities.
It is the second extension to the regional NSW lockdown, which came into effect at 5pm on August 14.
The lockdown was originally meant to end on August 22, but was extended by a week after cases continued to grow.
The two-week extension announced on Thursday means regional communities will be under the strict stay-at-home orders until at least 11.59pm September 10.
Meanwhile, greater Sydney will remain under lockdown until the end of September.
Two new Bathurst cases were also announced on Thursday, taking the city's total to 10, with one of those being a man with a Bathurst address who is in Sydney.
They were among the 35 recorded in the Western NSW Local Health District to 8pm Wednesday.
Residents are urged to come forward for testing.
In Bathurst, testing is available at Mount Panorama, Bathurst Base Hospital and the Bathurst Respiratory Clinic on the Charles Sturt University campus.
An additional testing site was opened at the Kelso Community Hub this week. It will operate from 9am to 3pm daily until Friday.
