Benalla officers were notified of the incident on Monday morning after footage was posted on Facebook.
Senior Sergeant Ash Mason said it was unclear where exactly the incident was filmed, but it appeared to have been somewhere on the Hume Highway.
A maroon and silver Holden VK Commodore performed the burnout for about a minute while being towed by a white van.
Two men are seen inside the car, with one squeezing out of the window, while a Mitsubishi Triton ute filled with tyres travels in the emergency lane.
Police are investigating possible links to the weekend's Winton Performance Car Mania event.
“It’s extremely disturbing and concerning behaviour which endangers themselves and other road users,” Senior Sergeant Mason said.
