In response to the Blues' 256, South Australia reached 7-197 at tea on day two of their Sheffield Shield clash after Cooper batted through the middle session.
The veteran batsman is unbeaten on 45 from 77 balls, while Tom Andrews, in contrast, has hit a quickfire 34 not out from 44 balls.
Cooper was the last recognised batsman remaining after SA were reduced to 5-128.
Paceman Trent Copeland has starred for the Blues in returning 4-53 from 22 tight overs with the Dukes ball.
The middle session was highlighted by the first wicket for debutant NSW leg-spinner Daniel Fallins (1-25) who had allrounder Joe Mennie caught and bowled.
NSW are seeking to bounce back from last weekend's defeat which toppled them from top spot on the Shield ladder.
